Translation initiation is a critical early step in the ‘life' cycle of positive-sense RNA viruses since production of viral proteins must precede genome replication. In Caliciviruses (of which noroviruses comprise an important genus), translation initiation depends on the VPg protein that is covalently attached to the 5´-end of the viral RNA. We have shown that norovirus VPg fulfils this role through direct association with a key component of the eukaryotic initiation machinery – eIF4G – and defined important structural details of this interaction. Intriguingly, our results suggest that the interaction can only occur once VPg has been proteolytically cleaved from the polyprotein precursor, which is the primary product of translation. In parallel investigations we have used NMR to examine the self-processing of this polyprotein by the viral NS6 protease that is initially embedded within it.

Stephen Curry教授现为英国帝国理工学院(Imperial College)生命科学系教授,于2012年当选皇家生物学会会士(Fellow, Royal Society of Biology),是世界知名的结构病毒学家。其研究聚焦在以小RNA病毒及诺如病毒为代表的正链RNA病毒的结构生物学方面。先后在Nature、Mol Cell、Nat Struct Mol Biol、PLoS Pathog、PNAS、JACS、J Virol、Nucleic Acid Res等期刊发表论文100余篇。此外,Curry教授目前也担任了多个组织(如英国Campaign for Science and Engineering等)的理事会成员或主席,并长期担任《卫报》专栏作家。
